Im Rahmen der vorliegenden Diplomarbeit wurde versucht, eine Methode zur An-bringung eines Fluoreszenzmarkers an Hexenuronsäure für analytische Zwecke zu entwickeln. Dazu wurden drei Reaktionstypen untersucht, mit deren Hilfe ein Reagens gezielt an die Doppelbindung einer Modellverbindung der HexA addiert werden sollte. Die angewendeten Reaktionen waren die Diels-Alder-Cycloaddition, die Aza-Michael-Addition und die radikalische Addition einer ungesättigten Carbonylverbindung nach vorhergegangener Hydroborierung des Alkens mit Catecholboran. Keine der untersuchten Reaktionen war jedoch geeignet, einen Substituenten an der ungesättigten Carboxylfunktion der HexA-Modellverbin-dungen zu verankern.<br />Weiters wurde untersucht, welche Reaktionsprodukte bei der Derivatisierung von Cellulose mit Ethylisocyanat (EIC) entstehen. Dabei konnte gezeigt werden, dass Cellulose bei der Umsetzung mit EIC am C-6 der Glucoseeinheit durch Bildung eines Urethans derivatisiert werden kann. Die weiteren freien Hydroxylgruppen an den Positionen 2 und 3 werden nicht umgesetzt. Im Gegensatz dazu reagiert Phenyl-isocyanat an allen drei OH-Gruppen pro Anhydroglucoseeinheit, bevorzugt aber an Position 3.<br />Nachdem imidazolbasierende ionische Flüssigkeiten zunehmend als Lösungsmittel für Zellstoffe eingesetzt werden, wurde untersucht, ob sich diese Verbindungen inert gegenüber der gelösten Cellulose verhalten. Anhand von 1-Butyl-3-methylimid-azoliumacetat (BMIM-OAc) konnte nachgewiesen werden, dass 1,3-disubstituierte Imidazoliumkationen mit Glucopyranose unter Adduktbildung reagieren. Weiters wurde die Reaktion der Cellulose am reduzierenden Ende mit BMIM-basierenden ionischen Flüssigkeiten unter Verwendung von Isotopenmarkierung, Fluoreszenz-markierung und Größenausschlusschromatographie bewiesen.<br />Damit wurde erstmals gezeigt, dass sich in Lösungen von Cellulose in BMIM-artigen IL weder Solvent noch Cellulose inert verhalten, sondern eine chemische Reaktion eingehen. Das hat wichtige Implikationen für die Verwendung ionischer Flüssigkeiten als Cellulose-lösungsmittel, insbesondere für medizinische Anwendungen.<br />

Within the present diploma thesis a method for the selective fluorescence labeling of hexenuronic acid for analytical purpose was to be developed. Therefore, three re-action types were tested to add a reagent to the double bond of model compounds of HexA, the Diels-Alder-cycloaddition, the aza-Michael-addition and the radical addition of an unsaturated carbonyl compound after hydroboration of the alkene. None of these reactions proved suitable for the intended purpose.<br />In addition, the products of the reaction between cellulose and ethylisocyanate were investigated. It was demonstrated that cellulose is substituted only at the C-6 of the anhydroglucose units by forming an urethane derivative. The hydroxyl groups in position 2 and 3 were not affected. In contrast to this result, the reaction with phenylisocyanate proceeds at all three OH groups per anhydroglucose unit.<br />Because of the increasing interest in imidazole-based ionic liquids as solvents for cellulose, investigations were performed to answer the question, if this class of solvents behaves inertly in cellulose solutions. It was proved that 1,3-disubstituted imidazolium cations react at C-2 with the reducing end of glucopyranoses under formation of a carbon-carbon bond. The analogous reaction of BMIM-based ionic liquids at the reducing end of celluloses was verified by means of isotopic labeling, fluorescence labeling and size-exclusion chromatography. It was shown that in solutions of cellulose in BMIM-type ionic liquids neither the solvent nor the cellulose are inert, but they undergo a chemical reaction instead. This fact has important implications on the use of ionic liquids as cellulose solvents.
