In der Seegras Spezies Posidonia oceanica wurde in einer Vorstudie die wertvolle bioaktive Cichoriensäure identifiziert. Eine Literatursuche zur Gewinnung von Phytochemikalien aus Pflanzen ergab eine Vielzahl von Möglichkeiten und Methoden wie: Mazeration, Wasserdampfdestillation, Soxhlet, Ultraschall assistierte, Pulsed Electric Field bzw. High Voltage Electrical Discharges, Super Critical Fluid, Pressurized Liquid, Mikrowellen assistierte Extraktion und die mechanische Direktpressung. Aufgrund der Instabilität der Cichoriensäure und der Vorgabe ein möglichst ökologisches Verfahren anzuwenden wurden systematische Versuchsserien der Ultraschall assistierten Extraktion, der Mazeration und der Direktpressung, kombiniert mit verschiedenen Vorbehandlungen um die aussichtsreichste und effizienteste Methode zu identifizieren, durchgeführt. Zur Bewertung der experimentell untersuchten Verfahren wurde die Ausbeute hinsichtlich der Cichoriensäure mittels HPLC quantitativ analysiert und parallel der Gesamtphenolgehalt für jede Versuchsvariante bestimmt. Es hat sich herausgestellt, dass durch die Ultraschallbehandlung eine wesentliche Verbesserung der Ausbeute (>200 % Cichoriensäuregehalt der einfachen Extraktion) erreicht wird. Daher könnten möglicherweise auch andere Zellaufschließende Verfahren (PEF/HVED) mit dem gleichen Resultat eingesetzt werden. Es konnte gezeigt werden, dass durch direktes Auspressen der Pflanzen ebenfalls eine Gewinnung der bioaktiven Substanzen mit einer ähnlichen Ausbeute des Gesamtphenolgehaltes erzielt wird. Voraussichtlich bringt die Kombination von einer Extraktionsmethode als Vorbehandlung mit anschließender Auspressung bzw. Mehrfachpressung mit oder ohne erneuter Lösungsmittelzugabe eine weitere Verbesserung der Ausbeute. In der feuchten Biomasse kommt es zu sehr raschen Abbauvorgängen v.a. der Cichoriensäure (56 % am ersten Tag) welche nur durch schonende Trocknung hintangehalten werden konnten. Auch in den Extrakten wurde eine Abnahme der Phytochemikalien mit der Zeit gemessen, allerdings mit einer wesentlich geringeren Geschwindigkeit (35 % in 70 Tagen). Es empfiehlt sich daher den Pflanzenrohstoff sowie die Extrakte rasch zu verwerten oder schonend zu trocknen.

Recent research on the seagrass species Posidonia oceanica showed the presence of the valuable bioactive compound cichoric acid in the plant. By doing a literature study on the isolation of phytochemicals from plants, a variety of possibilities and processes was found: maceration, steam distillation, Soxhlet, ultrasound assisted, pulsed electric field resp. high voltage electrical discharges, super critical fluid, pressurized liquid, microwave assisted extraction and mechanical pressing. Considering the instability and the thermally sensitive nature of cichoric acid as well as to fulfill the ecological demands, the most suiting approaches of ul-trasound assisted extraction, maceration and mechanical pressing were chosen for the test series. Different pretreatment processes were included to identify the most promising and efficient methods. For the evaluation of the experimentally investigated procedures the recovery of cichoric acid, measured quantitatively by HPLC, and of total phenolic com-pounds (FCR) was determined. The results have shown that the ultrasound assisted extraction had a substantial improvement on the yield (>200 % of cichoric acid content) compared to simple extraction. Considering this result, other cell disrupting applications (PEF and HVED) could lead to a similar output. The experiments also showed that by using mechanical pressing of the plant material the yielding of the total phenolic com-pounds nearly reaches the magnitude of the other approaches. It is most likely that a combination of an extraction method as pretreatment with following me-chanical pressing, possibly by multistep pressing with or without fresh solvent, would lead to a further improved yield of the phytochemicals. Noticeably rapid degradation processes, particularly of cichoric acid (56 % on the first day), were observed, which was only prevented by gently drying the plant material. Several measurements of the obtained extracts showed a much lower degradation rate of the phytochemicals (35 % in 70 days). A quick utilisa-tion or a gentle drying is recommended, as this will scale down the loss through degradation.
