EINLEITUNG:<br />Aufgrund seiner besonderen Eigenschaften, wie mechanische Belastbarkeit und hämostatische Wirkung, stellt Collagen ein wichtiges Biomaterial in zahlreichen Anwendungen im Bereich des Tissue Engineerings dar. In der vorliegenden Arbeit wird die Etablierung einer Methode zur Isolierung von Collagen aus humaner Placenta beschrieben. Derzeit wird Collagen hauptsächlich aus Gewebe tierischen Ursprungs gewonnen, was zu inflammatorischen und immunologischen Reaktionen führen kann. Daher wäre insbesonders im Bereich des Tissue Engineering der Einsatz von humanem Collagen anzustreben. Humane Placenta stellt eine ideale Quelle f¨ur die Collagenisolierung dar, da es in Spitälern als Abfallprodukt in größerer Menge und in gleichbleibender Qualität anfällt.<br />METHODEN:<br />Placenten wurden mit Pepsin enzymatisch verdaut und Collagen der Typen I, III, IV, und V mittels nachfolgenden wiederholten Salzfällungen und Dialyse isoliert. Die Charakterisierung der einzelnen Collagentypen erfolgte durch biochemische Analysemethoden, wie Aminosäurenanalyse und Western Blotting. Thermostabilität wurde mittels Circulardichroismus Spektrometrie untersucht. Untersuchungen hinsichtlich der antigenen Wirkung der isolierten Collagene erfolgte an mononukleären Zellen des peripheren Blutes (PBMC) durch Messung der Proliferation mittels BrdU-ELISA. Zytotoxizität wurde durch die Bestimmung der Viabilität von C2C12 Zellen mittels MTT-Assays untersucht. Hämostatische Wirkung der Collagene wurde mithilfe von potentiometrischen Messungen der Thrombocytenaggregation bestimmt.<br />RESULTATE:<br />Collagen der Typen I und III konnten in hohen Ausbeuten von über 200µg/100g Gewebe isoliert werden, Collagen Typ V nur in geringen Mengen. Collagen Typ IV konnte nur in Fragmenten isoliert werden, da seine globuläre Strukturen durch Pepsin angegriffen wurden. Analysen mittels MTT-Test zeigten, dass die isolierten Collagene keinerlei Zytotoxizität aufwiesen und sich daher für den Einsatz in der Zellkultur eignen. Ebenso zeigten die Ergebnisse der Untersuchungen bezüglich der antigenen Wirkung, dass die isolierten Collagene keinerlei immunologischen Reaktionen in PBMC in vitro auslösten. Weiters wiesen die Collagene hämostatische Wirkung und ausreichende Thermostabilität für medizinische Anwendungen, wie Wundverbände auf.<br />SCHLUSSFOLGERUNG:<br />Collagene konnten in zufriedenstellender Menge aus humaner Placenta isoliert werden. Die isolierten Collagene induzierten keinerlei zytotoxische oder immunologische Reaktionen. Humanes Collagen wäre ein ideales Material für zahlreiche Anwendungen in Biomedizin und Tissue Engineering.<br />

INTRODUCTION:<br />Collagen embodies an important biomaterial with numerous applications in tissue engineering due to its prominent features, such as mechanical strength and hemostatic activity. Here, we describe the establishment of an isolation method of collagen from human placenta. Currently, collagen is most often obtained from animal tissues, which may result in inflammatory or immunological reactions. Hence, collagen of human origin would be highly advantageous in the field of tissue engineering. Human placenta appears to be an ideal source of collagen, being a waste product in hospitals and available in consistent quality.<br />METHODS:<br />Collagen types I, III, IV and V have been isolated from human placenta by enzymatic digestion of placental tissue with pepsin and subsequent sequences of salt precipitations and buffer exchanges via dialysis.<br />Collagens were characterized by biochemical methods, i.e. amino acid analysis and western blotting. Thermostability was investigated by circular dicroism spectroscopy. Antigenicity of isolated collagens was tested in peripheral blood mononuclear cells (PBMCs) and influence on proliferation of PBMCs was determined by BrdU-ELISA. Cytotoxicity was tested on cultured C2C12 cells by determining viability via MTT-assay.<br />Hemostatic activity of collagens was determined by potentiometric measurements of platelet aggregation.<br />RESULTS:<br />Collagen types I and III could be isolated with a high yield of >200µg/100g wet tissue, whereas collagen type IV and V were obtained in low quantities. Collagen type IV could only be obtained in fragments since its partial globular structures were digested by pepsin. MTT-assay revealed that isolated collagens showed no cytotoxicity and can be used in cell culture applications. Results of antigenicity assays indicated that collagens do not induce an immunological reaction in PBMCs in vitro. Collagens exhibit hemostatic action and thermostability, shown to be sufficient for medical applications such as wound dressings.<br />CONCLUSION:<br />Collagen can be isolated in sufficient amounts from human placenta without inducing cytotoxic or immunological reactions. Human collagen may be an ideal material for various biomedical and tissue engineering applications.<br />
