Methoden des oxidativen biomimetischen Ligninabbaus, wobei Lignin-Abbaureaktionen ligninolytischer Enzyme simuliert werden - wurden erstmals zur Vorbehandlung von Weizen- (Triticum aestivum L.) und Haferstroh (Avena sativa L.) angewandt, um die Freisetzung reduzierender Zucker nach der enzymatischen Hydrolyse, sowie die Biogas Produktion zu erhöhen. Die Untersuchungen wurden mit verschiedenen Hydroperoxiden in Gegenwart von Übergangsmetallen - hauptsächlich Kupfer und Eisen - und Koordinatoren durchgeführt. Zu Beginn wurden verschiedene Pyridinderivate als Cu(II)-Komplexe in Verbindung mit Hydroperoxiden auf den Abbau von Veratrylalkohol untersucht. Terpyridin und die Kombination von Terpyridin und Bipyridyl eigneten sich am besten als Koordinatoren und spalteten Veratrylalkohol zu Veratraldehyd und Veratrylsäure - entsprechend dem Abbauschema von Ligninperoxidase. Im Gegensatz dazu resultierten nur undefinierte Produkte aus der Spaltung durch Fenton-basierte Reaktionen ohne Koordinator. Das koordinierte Kupfersystem baute auch die sehr resistenten synthetischen Farbstoffe Phenolrot und Azure B ab.<br />Die wirksamste Vorbehandlung von 1 cm - Weizenstroh erfolgte durch ein optimiertes 2-stufiges System bestehend aus EDTA und Cu/ tp/H2O2 bei pH 11,5, 90°C. Damit wurde der geringste minimale Ligningehalt von Weizenstroh (6% Restlignin bzw. 71% Ligninabbau) und der höchste Wert hydrolysierbarer Polysaccharide, ausgedrückt als reduzierende Zucker (704 mg red. Zucker / g Stroh bzw. 100% des theoretischen Maximalwertes) gemessen. Die Erniedrigung der Reaktionstemperatur auf 60°C zur Steigerung der Energieeffizienz bei gleichzeitiger Verringerung der Partikelgröße auf 2 mm führte bei der Vorbehandlung von Weizenstroh zu nur 68% Ligninabbau und 447 mg reduzierenden Zucker (67% theoretischer Maximalwert). Bei der Untersuchung des Biogaspotenzials, erreichte Cu/tp/H2O2 - vorbehandeltes Stroh die höchste Methanproduktivität. 100% des theoretischen Biogas Ertrages (basierend auf reiner Cellulose) wurde mit dem Feststoffanteil aus der EDTA-Cu/tp/H2O2 Vorbehandlung von Weizenstroh erreicht, was mit den entsprechenden Werten für reduzierende Zucker korreliert. Die Reduktion der Vorbehandlungstemperatur von 90°C auf 60°C verringerte auch die Methankonzentration deutlich.<br />In dieser Arbeit wurde erstmals gezeigt, dass oxidative biomimetische Systeme interessante Methoden sowohl zur Verbesserung der Zugänglichkeit der Holocellulose in Stroh für die enzymatische Hydrolyse als auch für die mikrobiellen Enzyme bei der anaeroben Umsetzung sind und das Biogaspotenzial von Stroh erhöhen. Weiters wurde eine indirekte Analysenmethode zur Vorhersage wichtiger Prozessparameter in Biogas Fermentationen entwickelt. Dazu wurden Fourier transformierte Nahinfrarotspektren (FT-NIR Spektren) des vorbehandelten Materials in Kombination mit chemometrischen Methoden verwendet. Der Masseverlust nach der Vorbehandlung, der Ligningehalt und der Anteil hydrolysierbarer Polysaccharide ausgedrückt als reduzierende Zucker kann mittels FT-NIR in Kombination mit uni- und multivariater Datenanalyse der Spektren in leistungsstarker Weise analysiert werden.<br />Aber auch die Biogas Produktivität aus dem vorbehandelten Stroh, die Gesamttrockensubstanz, sowie die organische Trockensubstanz des Schlamms konnten nur auf Basis der Substratqualität nach dem Vorbehandlungsschritt sehr genau bewertet werden. Diese Ergebnisse empfehlen die NIR Spektroskopie als eine alternative und kostengünstige Analysemethode für die Prozessanalytik bei der Biomethan Produktion, die robuste und validierte Vorhersagemodelle liefert.<br />

Methods of biomimetic oxidative lignin degradation, postulated to mimic the lignin biodegradation mechanism of ligninolytic enzymes in nature was first time examined for pre-treatment of wheat (Triticum aestivum L.) and oat (Avena sativa L.) straw with the aim of improving the release of reducing sugars after enzyme hydrolysis and increasing the biogas production. The studies were conducted with several hydroperoxides in the presence of catalyzing transition metal ions, mainly copper or iron, and coordinators. Screening studies of copper (II) and pyridine derivative complexes in the presence of hydroperoxides on veratryl alcohol degradation were carried out initially. It was found that the catalytic reaction of Cu(II)/tp-bpy/H2O2 as well as Cu(II)/tp/H2O2 performed best in degrading veratryl alcohol to corresponding veratraldehyde and veratric acid similar to the lignin peroxidase pathway. Contrary to that, only undefined products were observed from the degradation by Fenton-based reaction without complexing agent. The coordinated Cu- system also strongly degraded recalcitrant synthetic dyes, Phenol red and Azure B.<br />The most potential pre-treatment system of 1-cm wheat straw was an optimized 2-step EDTA Chelation-Cu(II)/tp/H2O2-system at pH 11.5, 90°C yielding the minimum residual lignin content of wheat straw (6 % residual lignin or 71% lignin removal) and the highest amount of hydrolysable polysaccharides expressed as reducing sugars (704 mg sugars/ g straw or ~100 % theoretical yield). Decreasing the temperature to 60°C for a more energy efficient process and the particle size to 2-mm for wheat straw pre-treatment, resulted in only 7% residual lignin (68% lignin removal) and 447 mg sugar/ g straw (67% theoretical yield).<br />For biogas potential test, treated straw by Cu(II)/tp/H2O2 achieved the highest methane productivity. 100% biogas theoretical yield (based on cellulose model compound) was achieved when the solid fraction from the optimized EDTA chelation-Cu(II)/tp/H2O2 system pretreatment of wheat straw was used as the substrate, correlating to the results of released reducing sugars. Decreasing the pretreatment temperature from 90°C to 60°C also considerably reduced methane concentration.<br />Thus, it was shown first time in this work, that oxidative biomimetic systems are interesting tools to improve the accessibility of holocellulose in straw for enzymatic hydrolysis and also for the microbial enzyme systems in anaerobic digestion, increasing the biogas potential of straw. An indirect analytical procedure to predict the important process parameters of biogas fermentations using Fourier transform near-infrared (FT-NIR) spectra of pre-treated material in combination with chemometrics was developed. Weight loss, residual lignin content and hydrolysable polysaccharides in terms of reducing sugars of the pre-treated straw can be powerfully analysed by Fourier transform near-infrared (FT-NIR) spectroscopy in combination with uni- and multivariate mathematical spectral data analysis. In addition, the assessment of biogas productivity from pre-treated straw and the parameters, total solids (TS) and volatile solids (VS) could be accurately evaluated based on substrate quality after pretreatment. The findings in this work suggest NIR spectroscopy as an alternative and low-budget analytical tool for process analytical chemistry (PAC) of lignocellulose-to-engery conversion in biomethane production providing robust and validated prediction models.
