Abstract:
Mikroalgen sind potenzielle Rohstoffe für die nachhaltige Herstellung einer Vielzahl von Produkten. Ihre biotechnologische Kultivierung ist jedoch wegen der unzureichenden und ungleichmäßigen Lichtverteilung in Photobioreaktoren (PBR) unwirtschaftlich. Daher sind niedrige volumetrische Biomassekonzentrationen zu erwarten, die zu geringeren Gesamtproduktivitäten führen. Um diese Hindernisse zu umgehen, wurden folgende Strategien zur biotechnologischen Prozessintensivierung untersucht: (i) Natürliche Lebensräume wurden erforscht und die entdeckten Arten als potenzielle neue Produktionsstämme charakterisiert. (ii) Die Kulturen wurden mit organischen Kohlenstoffquellen für das mixotrophe Wachstum versorgt. (iii) Molekularbiologische Werkzeuge wurden für das Metabolic Engineering eingesetzt. (iv) Eine neuartige prozessanalytische Technologie (PAT) wurde für die Inline-Produktüberwachung in PBR untersucht. Diese Ansätze wurden am Beispiel der beiden Produkte Carotinoide und Polyhydroxyalkanoate untersucht. Zwei Stämme, die in einem Gletschervorland in Island gesammelt und als Tetraedron minimum und Coelastrella terrestris identifiziert wurden, wurden auf die Produktion von Sekundärcarotinoiden (SC) untersucht. Für T. minimum und seine Familie Hydrodictyaceae war dies der erste Bericht über den SC-Stoffwechsel in der Literatur. In mixotrophen C. terrestris-Kulturen war die Konzentration des seltenen SC Adonixanthin bemerkenswert hoch für Grünalgen, sodass ein biotechnologischer Produktionsprozess vorgeschlagen wurde. Außerdem wurde die Cyanobakterienmutante Mt_a24, die das Biopolymer Polyhydroxybutyrat (PHB) akkumuliert, gentechnisch verändert, indem das Kohlenhydratstoffwechselgen exoD ausgeschaltet wurde. Dadurch erhöhte sich die intrazelluläre PHB-Menge im Vergleich zur ursprünglichen Mutante. Darüber hinaus wurde ein in situ PHB-Quantifizierungs-PAT-Tool für die Echtzeit-Produktüberwachung während der PBR-Kultivierung entwickelt, das es ermöglicht, den optimalen Erntezeitpunkt zu bestimmen und auf eventuelle Prozessabweichungen zu reagieren.

Microalgae are potential resources for the sustainable production of a great variety of products. However, their biotechnological cultivation is uneconomic mostly due to insufficient and inconsistent light distribution in photobioreactors (PBR). Hence, low volumetric biomass concentrations leading to reduced overall productivities can be expected. To circumvent these obstacles, following strategies were investigated for biotechnological process intensification: (i) natural environments were explored and the discovered species characterized as potential novel production strains; (ii) organic carbon sources were provided to cultures for mixotrophic growth; (iii) molecular biology tools were utilized for metabolic engineering; and (iv) a novel process analytical technology (PAT) was examined for in-line product monitoring in PBR. These approaches were investigated by the example of the two products, carotenoids and polyhydroxyalkanoates. Two strains collected in a glacier foreland in Iceland identified as Tetraedron minimum and Coelastrella terrestris were analyzed for secondary carotenoid (SC) production. For T. minimum and its family Hydrodictyaceae this was the first report of SC metabolism in literature. In mixotrophic C. terrestris cultures the concentration of the rare SC adonixanthin was remarkably high for green algae, thus, a biotechnological production process was proposed. Further, the cyanobacteria mutant Mt_a24 accumulating the biopolymer polyhydroxybutyrate (PHB) was genetically engineered by knocking out the carbohydrate metabolism gene exoD. This increased the intracellular PHB amounts compared to the parental mutant. In addition, an in situ PHB quantification PAT tool for real-time product monitoring during PBR cultivations was developed which made it possible to determine the optimum point of harvest and to react to eventual process deviations.
