Adaptive laboratory evolution experiments with a hypermutator strain of thermoanaerobacter kivui

Abstract

Thermoanaerobacter kivui ist ein anaerobes, thermophiles Bakterium mit einer optimalen Wachstumstemperatur von 66 °C und bietet Energieeinsparungen für nachhaltige Bioprozesse an. Unter Verwendung eines endogenen CRISPR/Cas-Systems mit einer Geneditierungseffizienz von 39 %, wurde eine Punktmutation in die DNA-Polymerase III-α Typ PolC eingeführt, die eine hochkonservierte Domäne und für die DNA-Replikation verantwortlich ist. Unerwartet, nach 30 seriellen Transfers von PolCC628Y und dem Wildtyp- Stamm reduzierte sich die Zahl der Mutationen. Die Untersuchung der einzigartigen Mutationen in den Proben nach 30 Transfers hatte der Wildtyp-Stamm eine 1,5- bis 2,9-fach höhere Mutationsrate als der PolCC628Y-Stamm. Die Punktmutation in der Polymerase- und Histidinol-Phosphatase-Domäne könnte die funktionelle und strukturelle Integrität der DNA- Polymerase III vom PolC-Typ beeinträchtigen und sie anfälliger für höhere Temperaturen gemacht haben. Außerdem wurden Experimente mit gerichteter Evolution mit Formiat als Substrat durchgeführt. Dabei wurden Wildtyp und ΔhydA2 Stamm, welchem eine Hydrogenase fehlt, die verantwortlich für die Produktion von Formiat aus H2 und CO2 ist, untersucht. Diese Experimente lieferten einen Proof-of-Concept für künftige ALE-Experimente mit mixotrophen Bedingungen und zeigten ein langsameres Wachstum des ΔhydA2-Stammes.In weiterer Folge könnte man den Stamm, der in dieser Arbeit entwickelt wurde, für die Entwicklung von biotechnologischen Anwendungen einsetzen.

Thermoanaerobacter kivui is an anaerobic, thermophilic bacterium with an optimal growth temperature of 66 °C and may offer considerable energy savings for sustainable bioprocesses. Using an endogenous CRISPR/Cas system with an editing efficiency of 39 %, a single point mutation was introduced in the PolC-type DNA Polymerase III-α, a highly conserved domain essential for DNA replication. Unexpectedly, after 30 serial transfers of PolCC628Y and wild-type strain, the total mutation decreased in all samples. After normalizing, the wild-type strain had a 1.5- to 2.9-fold higher mutation rate than the PolCC628Y strain. The point mutation in the Polymerase and Histidinol Phosphatase domain might have interfered with the functional and structural integrity of PolC-type DNA Polymerase III and made it less viable at higher temperatures. Furthermore, ALE experiments with the growth on formate were conducted to investigate wild-type and ΔhydA2 strain, a strain missing a hydrogenase responsible for creating formate out of CO2 and H2. These experiments provided proof of concept for future ALE experiments with mixotrophic conditions and showed formate dependency of the ΔhydA2 strain.This work investigates potential biotechnological applications for T. kivui, accelerating the establishment of a circular carbon economy.
