Simultaneous in-line Raman and mid-IR spectroscopy for monitoring of complex crystallization systems in the pharmaceutical industry

Abstract

Die Kristallisation ist ein wichtiges Verfahren für die Trennung und Reinigung von Reaktionsprodukten in Industriezweigen wie der Biochemie, der Feinchemie oder der Pharmazie. Kristallisationsprozesse können mehrere Polymorphe erzeugen, von denen normalerweise nur eine die gewünschten physikalischen (Löslichkeit, Stabilität, Schmelzpunkt usw.) und chemischen (Reaktivität) Eigenschaften aufweist. Die Bildung unerwünschter polymorpher Formen beeinträchtigt die Produktqualität und damit die Eignung für den vorgesehenen Verwendungszweck erheblich. Daher ist eine sorgfältige Überwachung des Kristallisationsprozesses von entscheidender Bedeutung, um die Polymorphbildung und die Gesamtprozesszeit (Endpunkt) zu kontrollieren, was letztendlich zu effizienteren und optimierten Produktionszyklen führt.Raman und mittlere Infrarot Spektroskopie dienen als zerstörungsfreie Echtzeit-Überwachungs-instrumente für Kristallisationsprozesse. In dieser Arbeit wird die Ableitung von Konzentrationsprofilen von Produkt, Lösungsmittel und Antisolvens, basierend auf uni- und multivariater Datenanalyse aufgezeigt, die eine detaillierte Untersuchung und ein besseres Prozessverständnis ermöglichen. Während die Information in den Raman-Spektren von der kristallinen Phase dominiert wird, was ein Polymorph-Screening ermöglicht, werden in der FTIR-ATR-Konfiguration hauptsächlich Signale der gelösten Produktphase detektiert. Um ein stabiles Endpunktkriterium für Kristallisationsprozesse zu erhalten, wurden mehrere Methoden zur Endpunktdetektion untersucht. Der Einfluss der Temperatur muss bei der Analyse ebenfalls berücksichtigt werden, da diese ein entscheidender Faktor für Kristallisationsprozesse ist, die auch die beiden schwingungsspektroskopischen Techniken durch Signaländerungen in Form von Bandenverschiebungen, Verbreiterungen oder Intensitätsänderungen beeinflussen. Für zeitaufgelöste Messungen von schnellen Kristallisationsprozessen wurde ein kontinuierlicher Durchflussreaktor verwendet, um polymorphe Übergangszustände bei unterschiedlichen Kristallreifungszeiten zu identifizieren.Die Kombination von Raman- und FTIR-Inline-Spektroskopie ermöglicht die Echtzeitüberwachung des Reaktionsverlaufs, gewährt Einblicke in die Auswirkungen wichtiger Prozessparameter und sichert die Produktqualität durch die Bestätigung der polymorphen Reinheit. Die polymorphe Reinheit der Produkte konnte für beide untersuchten Spezies bestätigt werden, da keine polymorphen Übergänge festgestellt wurden. Mithilfe semi-quantitativer Daten, die mit den PAT-Tools erfasst wurden, wurde eine Methode zur Endpunkterkennung entwickelt, die auf stabilisierenden Trends in Batch-Kristallisationsexperimenten basiert. Zudem wurde ein Leitfaden vorgeschlagen, der einen systematischen Arbeitsablauf für die spektroskopische Untersuchung von Kristallisationsreaktionen definiert. Dies bildet die Grundlage für die Entwicklung eines datengestützten Modells zur optimierten Prozesskontrolle in einem pharmazeutischen Produktionsprozess.

Crystallization is an important unit operation for separation and purification of many industrial products such as biochemicals, fine chemicals or pharmaceuticals. Crystallization processes may yield several polymorphic forms while typically only one form exhibits the desired physical (solubility, stability, melting point, etc.) and chemical (reactivity) properties. Production of the undesired polymorph(s) significantly impacts the product quality and hence, its suitability for the intended use. Consequently, careful monitoring of the crystallization process is crucial to control polymorph formation and overall process time (endpoint) ultimately resulting in more efficient production cycles.Raman and mid-infrared spectroscopy serve as a non-destructive, real-time monitoring techniques for crystallization processes. In this work, the extraction of concentration profiles of product, solvent and antisolvent, based on uni- and multivariate analysis, is shown allowing for detailed investigation and improved process understanding. While the information in the Raman spectra is dominated by the solid phase allowing for polymorph screening, the FTIR-ATR configuration predominantly detects spectra of the liquid phase. Several methods for endpoint detection of crystallization processes were investigated to achieve a stable criterion. Also, the effect of temperature needs to be considered for analysis as it is a pivotal factor for crystallization processes affecting both vibrational spectroscopy techniques through alterations in terms of band shifts, broadening or intensity. To enable time-resolved monitoring of fast crystallization processes, a flow reactor design was employed to detect polymorphic transition states at varying crystal maturation times.Combining Raman and FTIR in-line spectroscopy enables real-time monitoring of reaction progression, provides insights into the effects of key process parameters, and ensures product quality by confirming polymorphic purity. The polymorphic purity of the products was confirmed for both investigated species, as no polymorphic transitions were detected. A method for endpoint detection based on stabilizing trends in batch crystallization experiments was developed using the semi-quantitative data collected by the PAT tools. Additionally, an instructional guideline was proposed, outlining a systematic workflow for the spectroscopic investigation of crystallization reactions. This lays the foundation to establish a data-driven model for optimized process control in a pharmaceutical production process.
