Abstract:
Diese Diplomarbeit beschreibt einen neuen Algorithmus für die Berechnung von stufenweise adjustierten Konfidenzintervallen für adaptive gruppensequentielle Verfahren, vorgeschlagen in Brannath, Mehta und Posch (2008). Die Grundidee ist, basieren auf der Methode von Müller und Schäfer (2001), die gesamte Fehlerwahrscheinlichkeit erster Art nach einer möglichen Designadaption zu bewahren, indem die Fehlerwahrscheinlichkeit des restlichen Designs auf die bis dahin beobachteten Daten bedingt wird. Wir geben in dieser Arbeit einen Überblick über adaptive gruppensequentielle Verfahren, die Berechnung von p-Werten und Konfidenzintervallen. Das Hauptaugenmerk liegt auf stufenweise adjustierten Konfidenzintervallen, entwickelt von Tsiatis, Rosner und Mehta (1984). Diese Konfidenzintervalle garantieren exakte Überdeckungswahrscheinlichkeit, falls keine Adaption vorgenommen wird.<br />Im Fall einer Adaption haben diese Konfidenzintervalle nur eine (schwach) konservative Überdeckungswahrscheinlichkeit. Der Algorithmus für die Berechnung der stufenweise adjustierten Konfidenzintervalle wird im Detail beschrieben und ist in C implementiert. Wir werden mit Hilfe von Simulationen zeigen, dass stufenweise adjustierte Konfidenzintervalle eine theoretisch exakte Überdeckungswahrscheinlichkeit haben und dass die berechneten Punktschätzer für den Behandlungseffekt praktisch erwartungstreu sind.<br />

This thesis describes a new algorithm for the calculation of stage-wise adjusted confidence intervals for adaptive group sequential trials proposed in Brannath, Mehta and Posch (2007). The key idea, based on the method of Müller and Schäfer (2001), is to preserve the overall type I error rate after a possible design adaptation, by preserving the null conditional rejection probability of the remainder of the trial at each time of an adaptive change. We will give an overview of adaptive group sequential designs, the calculation of p-values and confidence intervals. The main focus will be on stage-wise adjusted confidence intervals proposed by Tsiatis, Rosner and Mehta (1984). These confidence intervals provide exact coverage in the case of no adaptive changes. In the case of adaptive changes we can say that the calculated confidence interval is conservative. The algorithm for the calculation of stage-wise adjusted confidence intervals is described in detail and an example for the full implementation in C is given. We will show by simulations that the stage-wise adjusted confidence intervals provide exact coverage probability and the calculated point estimates for the treatment difference are median unbiased.
