Abstract:
Der Schwerpunkt dieser Arbeit lag darin eine Plattform zur Aufreinigung von Impfstoffen und verschiedenen Formulierungen, die auf intakten Viren basieren, zu entwickeln und diese mit Hilfe neuer analytischer Methoden zu charakterisieren. Eine Strategie zur Abtrennung von intakten Viruspartikeln aus den Impfstoffen von Hilfsstoffen wie Proteinen, Zucker und Salze wurde entwickelt, die notwendig ist für die weitere Analyse. Viruspartikelgröße und -verteilung von gereinigten Fraktionen wurde mit einem Nano-electrospray gas-phase electrophoretic mobility macromolecular analyzer (GEMMA) gemessen. Dieses Instrument liefert Informationen über die elektrophoretische Mobilität (Teilchendurchmesser) der einfach geladenen Partikel. Gemessene Partikeldurchmesser der Impfstoffproben wurden anschließend mit Messungen der Rasterkraftmikroskopie korreliert und mit Ergebnissen der Transmissionselektronenmikroskopie verglichen. Zusätzliche Informationen über die verschiedenen Vakzinpräparationen wurden durch Methoden wie SDS-PAGE, Elektrospray-Ionisations (ESI)-Massenspektrometrie und immunologische Tests gewonnen. Es konnte gezeigt werden, dass Größenausschlusschromatographie eine geeignete Methode ist, um die viralen Proben für die nachfolgenden Analysen vorzubereiten. Der elektrophoretische Durchmesser von Viruspartikeln wie dem Frühsommer-Meningoenzephalitis-Virus (FSME), Ross River Virus (RRV) und West Nile Virus (WNV) konnte mit Hilfe des GEMMA-Instruments gemessen werden. Für FSME Partikel beträgt der Durchmesser 46.8 ± 1.1 nm, für RRV 57.8 ± 0.8 nm und für WNV 44.3 ± 0.4 nm. Zusätzlich wurden die Partikelgrößen von humanen Papillomaviren, die aus einem kommerziell erhältlichen Impfstoff gewonnen wurden und von humanen Rhinoviren bestimmt. Mit Hilfe der ESI-Massenspektrometrie wurde auch das Molekulargewicht viraler Partikel, wie virus like particles bestimmt und verglichen mit NanoES-GEMMA Daten. Des Weiteren wurde die Stöchiometrie von Antikörperfragmenten (Fab), die an virale Partikel binden, mittels NanoES-GEMMA-Spektren bestimmt und mit ESI-Massenspektrometrie Daten verglichen.
The focus of this work was to establish a platform to purify intact virus-based vaccines and their formulations for the subsequent characterization. New analytical methods should be established within a platform for their characterization. An analytical strategy for isolating intact vaccine particles from agents like proteins, sugars and salts was developed which is necessary for further vaccine analysis. Virus particle size and size distribution were measured with a nano-electrospray gas-phase electrophoretic mobility macromolecular analyzer (GEMMA), revealing information on electrophoretic gas-phase mobilities (particle diameter). Determined particle diameters were subsequently correlated to atomic force microscopy measurements and combined with results from transmission electron microscopy. Additional information about different vaccine preparations was gained by methods like SDS-PAGE, electrospray ionization (ESI) mass spectrometry and immunological tests. It turned out that size exclusion chromatography is a reliable method for viral sample preparation for further particle analysis. The electrophoretic diameters of viral particles of tick borne encephalitis virus (TBEV), Ross River virus (RRV) and West Nile virus (WNV) are determined by means of the GEMMA instrument, revealing diameters of 46.8 ± 1.1 nm for TBEV, 57.8 ± 0.8 nm for RRV and 44.3 ± 0.4 nm for WNV. In addition, a particle size estimation of human papilloma virus particles, purified from a commercially available vaccine, and human rhino virus particles could be performed. The molecular weight of viral particles were also investigated and corroborated by means of mass spectrometry and the stoichiometry of antibody fragments (Fab) that bind to viral particles was calculated from GEMMA spectra.
