Unravelling the interconnection between heterotrimeric G-protein signaling and light response in Trichoderma reesei (Hypocrea jecorina)

Abstract

In dieser Arbeit wurden Komponenten des Lichtsignal-Übertragungsweges im Zellulose abbauenden filamentösen Pilz Trichoderma reesei (Hypocrea jecorina) untersucht. Die Zellulose abbauenden Proteine dieses Pilzes, Zellulasen, werden in zahlreichen industriellen Prozessen verwendet, beispielsweise in der Bioethanolproduktion, die immer noch hohe Produktionskosten aufweist. Da der Einfluss von Licht zu einer Verdopplung der Zellulasetranskription in T. reesei führt, ist es von großer Bedeutung den Regulationsmechanismus der Lichtübertragung zu untersuchen und zu verstehen.<br />Während dieser Arbeit untersuchte ich insbesondere die Rolle des Lichtsignalregelproteins ENVOY im G-Protein-Signalweg, von dem bereits ein Einfluss auf die Zellulaseproduktion bekannt ist. G-Proteine sind Übermittler von extrazellulären Signalen zu intrazellulären Zielen. Die Transkription der G-Protein-Untereinheiten ist direkt reguliert von zwei Faktoren: ENVOY und PhLP1 (Phosducin-ähnliches Protein 1). Der Effekt von Licht ist zumindest teilweise übermittelt durch die zwei Blaulicht-Rezeptor-Proteine BLR1 und BLR2 zu ENVOY und dann weiter zu den G-Proteinen durch ENVOY und PhLP1. ENVOY kontrolliert die frühen Signalübertragungsvorgänge und führt dabei zu einer Reduktion der Transkription von PhLP1 und den G-Protein Untereinheiten. Bei längerer Beleuchtung regeln ENVOY und PhLP1 ihre eigene Transkription gegenseitig und erhalten so eine abgestimmte Menge ihrer Transkripte abhängig vom Lichtzustand. Der Licht abhängige Signalweg hat Auswirkungen auf fast alle Kohlenstoff abbauenden Glycosidhydrolasen. Dies bestätigt die Bedeutung von Licht auf den Zellulase Signalweg. Aus diesen Daten wurde ein Modell entwickelt, wie die Komponenten des Lichtsignalweges untereinander reguliert werden und eine mögliche Erklärung gefunden, wie das Lichtsignal zum Zellulasepromotor übertragen wird.

The aim of my PhD thesis was to analyze components of the light signaling cascade in the most prominent cellulose degrading filamentous fungus Trichoderma reesei (Hypocrea jecorina.). The cellulose degrading proteins of this fungus, cellulases, are used in many industrial applications such as bioethanol production, which is still very expensive. Since light doubles the amount of cellulase transcripts in T.<br />reesei it is very important to investigate and understand the regulation mechanisms behind the light signaling cascade.<br />In the course of this thesis I investigated in particular the role of a light regulatory protein, ENVOY, in the G protein signaling pathway, which is known to influence cellulase production. G proteins are mediators of extracellular signals to intracellular targets. The transcription of these G protein subunits is directly regulated by two factors: ENVOY and PhLP1 (phosducin like protein 1). The effect of light is at least partly transduced by the two blue light receptor proteins BLR1 and BLR2 to ENVOY and further transferred to the G proteins by ENVOY and PhLP1. ENVOY controls early signaling events by downregulation of PhLP1 and the G protein subunits. Upon prolonged illumination, ENVOY and PhLP1 are in a regulatory relationship and are assumed to adjust their amount of transcript levels at steady state levels dependent on the light status. The light dependent signaling pathway has impact on almost all carbohydrate degrading glycoside hydrolases, confirming the importance of light in the cellulase signaling pathway. I propose a model for the regulation of the components of the light signaling cacade and a possible explanation for the transduction of the light signal to the cellulase promotors.<br />
