The biocontrol agent trichoderma atroviride: role of G protein signalling in host recognition and analysis of its secondary metabolites

Abstract

Der filamentöse Pilz Trichoderma atroviride ist ein potenter Biokontrollorganismus und stellt eine gute Alternative als natürliches Fungizid für chemische Mittel dar. Als Mykoparasit befällt er pflanzenpathogene Pilze wie Botrytis cinerea, Fusarium spp., Rhizoctonia solani und Sclerotinia sclerotiorum. Der Vorgang des Mycoparasitismus gliedert sich in chemotrophes Wachstum, Wirtserkennung, Ausbildung von Infektionsstrukturen und Bildung von hydrolytischen Enzymen und antifungaler Metabolite. Um den Wirtspilz zu erkennen benötigt T.<br />atroviride eine Reihe von Rezeptoren an seiner Zelloberfläche. Bestimmte Rezeptoren sind an G-Proteine gekoppelt und leiten Signale von der Zelloberfläche an diese G-Proteine weiter und lösen damit eine Reihe von intrazellulären Signaltransduktionskaskaden aus. Im Rahmen dieser Dissertation wurde im Genom von T. atroviride nach allen G-Protein gekoppelten Rezeptoren (GPCRs) gesucht. Die gefundenen GPCRs wurden danach je nach ihren Eigenschaften in acht verschiedene Klassen unterteilt. Das Stilllegen oder silencen des gpr1 Gens führte zu einer T. atroviride Mutante mit stark verlangsamtem Wachstum. In Konfrontation mit dem Pflanzenpathogen Rhizoctonia solani zeigte die Mutante ein komplett avirulentes Verhalten und konnte den Wirt nicht mehr überwachsen oder lysieren. Für den Mykoparasitismus typische Infektionsstrukturen wurden nicht mehr ausgebildet, welche jedoch durch Zugabe von cAMP ins Medium wieder hergestellt werden konnten. Die Expression von Genen die für lytische Enzyme kodieren war in der Mutante während der Konfrontation mit dem Wirtspilz nahezu nicht vorhanden. Im Gegensatz dazu konnte gezeigt werden, dass die Enzymaktivität der Endo- und Exochitinasen in der Mutante im Vergleich zum Wildtyp deutlich erhöht war, was sich auch in der Transkription der für die Chitinasen kodierenden Gene reflektiert. Weiters wurde eine Methode entwickelt um die Produktion antifungaler Metaboliten im Wildtyp von T. atroviride zu untersuchen.

The filamentous fungus Trichoderma atroviride is a potent biocontrol organism and exhibits a good alternative as a fungizide compared to chemicals. As a mycoparasite it attacks plant pathogenic fungi like Botrytis cinerea, Fusarium spp., Rhizoctonia solani and Sclerotinia sclerotiorum. Mycoparasitism is divided into different parts like chemotrophic growth, recognition of the host, infectionstructure formation and secretion of lytic enzymes and antifungal metabolites. For host recognition T. atroviride possesses a bunch of different receptors on its cell surface. Some of them are connected with G proteins and transmit signals from the cell surface to these G proteins to trigger intracellular signalling cascades. In this thesis all possible G protein coupled receptors(GPCRs)have been found in the genome of T. atroviride.<br />All of the GPCRs are divided into eight different classes conferring their abilities. Silencing of the gene gpr1 leads to a T. atroviride mutant with reduced growth. In confrontation with the plant pathogen R.<br />solani the mutant was completely avirulant and unable to parasitize the host. Mycoparasitism related infection structure formation was stopped but could be restored after adding some endogenous cAMP. Expression of genes coding for lytic enzymes was completely downregulated in the mutant during confrontation. The enzymatic activity of exo- and endochitinases of the mutant was significantly elevated compared to the wild-type confirming the elevated transcription of the chitinase encoding genes. Furthermore a LC/MS-MS based method was used to determine the production of antifungal metabolites by T. atroviride.
