Cost-to-Go Model Predictive Control for Enhanced Optimization of Bioprocesses

Abstract

Die Verwendung von Fermentationen in Bioreaktoren zur effizienten Produktion komplexer Moleküle hat in vielen Bereichen an Bedeutung gewonnen und dient der Substitution aufwendiger chemisch-technologischer Prozesse. Sowohl die qualitativen als auch die quantitativen Anforderungen an das vom Mikroorganismus produzierten Endprodukt sind hoch. Das Ziel ist somit ein in jeder Hinsicht optimiertes Prozessergebnis zur Erfüllung spezifischer Vorschriften bezüglich Reinheit und Prozessgestaltung als auch der kostengünstigen Auslegung der Aufreinigung im Downstream. Zur Erfüllung eben jener Anforderungen ist die Prozessführung und damit auch die hoch entwickelte Steuerung und Regelung solcher Fermentationen essentiell. Daher wird in dieser Arbeit ein neuartiger, modellbasierter zweistufiger Ansatz zur optimalen Regelung von Bioprozessen behandelt. Kennzeichnend für diesen Ansatz ist die Einbindung der optimalen cost-to-go. Die cost-to-go repräsentiert die minimalen Kosten, die zwischen einem beliebigen Zustandspunkt zu einem bestimmten Zeitpunkt und dem angestrebten Endzustand benötigt werden, um einen Prozess mit seinen Systemeingängen entlang einer optimalen Trajektorie zu führen. Zuerst wird Dynamic Programming (DP) für die offline Berechnung der optimierten cost-to-go auf Basis eines nichtlinearen Zustandsraummodells des physikalisch beschränkten Bioprozesses verwendet. Nach erfolgter Optimierung wird die cost-to-go für sämtliche Zustandspunkte über die Prozesszeit in einer mehrdimensionalen Matrix abgelegt und in den Algorithmus eines modellprädiktiven Reglers (MPC)integriert. Im Gegensatz zum klassischen MPC-Ansatz, bei dem zur Stellgrößenermittlung der Fehler zwischen einer prädizierten Ausgangsgröße und einer Referenztrajektorie innerhalb des Prädiktionshorizonts minimiert wird, gewährleistet anschließend der cost-to-go MPC, dass sich der Zustand am prädizierten Horizont aufgrund der Berücksichtigung von Terminal Costs stets auf einer cost-to-go befindet. So kann der Prozess bei abweichendem Verhalten, zum Beispiel durch Störungen und Modellierungsungenauigkeiten, einen definierten Endzustand optimal erreichen. Die Performance dieses Ansatzes wird abschließend durch simulative Fallstudien, in denen die Regelungsstrategie anhand einiger Szenarien validiert wird, gestützt und mit dem klassischen Ansatz zur modellprädiktiven Regelung verglichen.

The use of fermentations in bioreactors for the efficient production of complex molecules has gained importance in many areas and serves as a substitute for complex chemical-technological processes. Both the qualitative and quantitative requirements for the final product produced by the microorganism are high. The objective is therefore a process that is optimised in every respect to meet specific regulations regarding purity and process design, in addition to the cost-effective design of downstream purification. To fulfil these requirements, a sophisticated process control of such fermentations is essential. This thesis proposes a novel model-based two stage approach for the optimal control of bioprocesses. This method is characterised by the integration of the optimal cost-to-go which represents the minimum cost required to move from any state at any point in time to the desired final state along the optimal path. First, Dynamic Programming (DP) is used for the offline calculation of the optimised cost-to-go based on the nonlinear state-space model of the physically constrained bioprocess. After optimisation, the cost-to-go for all state points over the process time is stored in a multidimensional matrix and integrated into the algorithm of a Model Predictive Control (MPC). In contrast to the classic MPC approach, in which the error between a predicted output variable and a reference trajectory within the prediction horizon is minimised to obtain the actuating variable, the cost-to-go MPC ensures that the state at the predicted horizon is always on a cost-to-go due to the consideration of terminal costs. This allows the process to optimally reach a defined final state in the event of deviating behaviour, for example due to disturbances and plant-model mismatch. Finally, the performance of this approach is supported by simulative case studies in which the control strategy is validated using several scenarios and compared with the classic MPC approach.
