Synthesis, medicinal chemistry and anti-bacterial profiling of anthraquinone-derivatives towards multi-resistant bacteria starting from sltersolanol M as lead structure

Abstract

Ein neues Molekül aus der Altersolanol Familie wurde entdeckt. Da dieses Molekül gute antimikrobielle Aktivitäten zeigte wurde ein Syntheseplan entworfen. Krohn et al. (1988) hat einen Total- Synthese Plan von Altersolanol A publiziert. Wir haben anhand seiner Publikation einen alternativen Syntheseplan mit besserer Regioselektivität und somit besseren Ausbeuten entworfen. Weiters wurde die Struktur vereinfacht und die Targetmoleküle MG009 und MG029 erhalten. Diese beiden Substanzen zeigten gute Hemmwirkung auf gram positive Bakterien. MG009 zeigte beim MHK- Test eine hemmende Wirkung gegen MRSA, bei einer Konzentration von 0.49µg/mL und MG029 bei 0.98µg/mL. Ebenfalls wurde ihre Bakterizide Aktivität bestätigt. Beide Substanzen zeigten die besten Ergebnisse im Zytotoxizität Test, vor allem im Hautmodell zeigte MG009 eine noch weiter reduzierte Toxizität. Eine weitere wichtige Untersuchung war die Bildung von reactive oxygen species (ROS), welche bei beiden Molekülen ausgeschlossen werden konnte. Es konnte die Interkalation in die Zelluläre DNA ausgeschlossen werden. Mittels TOPO I Interkalations Test konnte bei geringen Konzentrationen keine Interkalation festgestellt werden. Bei höheren Konzentrationen zeigt MG009 interkalierende Aktivität. Dies war Anlass um dessen Anti- Krebs Eigenschaften zu untersuchen. Dafür wurde MG009 in einem ersten Test auf Krebszellen getestet und zeigte gute Ergebnisse, welche für weitere Studien verwendet werden kann.

A new compound of the altersolanol family was discovered. Due to its antimicrobial activity a synthesis plan was acquired. There exists a total synthesis plan for Altersolanol A described by Krohn et al. (1988) and an alternative synthesis way with better yields was established. Further the structure was simplified to get the target molecules MG009 and MG029. The two lead compounds showed good inhibition effects on the growth of gram positive bacteria. MG009 showed a MIC of 0.49µg/ml on MRSA and MG029 had a MIC of 0.98µg/ml on MRSA. Also their antibactericidal activity was approved. Both substances showed the best results in cytotoxicity assays especially on skin equivalent models MG009 showed a reduced toxicity. A very important investigation was the formation of reactive oxygen species (ROS) which could be excluded for MG009 and MG029 in our assay. We could also rule out the intercalation of MG009 and MG029 into the cell DNA. Via TOPO I assay the non intercalating tendencies with low concentrations was also confirmed. At higher concentrations MG009 showed intercalating tendencies. Due to this fact its anticancerous activity was tested in an first assay on different cancerous cells and showed good results which can be used for further investigations.
