Abstract:
Zwei-Photonen Polymerisation (2PP) ermöglicht die Herstellung von drei-dimensionalen Strukturen mit einer hohen Auflösung von bis zu 100nm.<br />Jedoch ist die Schreibgeschwindigkeit von konventionellen Systemen für großflächige Strukturen mit 10mm/s gering, was zu langen Bauzeiten führt.<br />Die Verwendung eines Galvoscanners ermöglicht höhere Schreibgeschwindigkeiten von bis zu 500mm/s; die maximale laterale Größe der Struktur ist jedoch durch das Sehfeld des Objektives beschränkt. Deshalb wird das Sehfeld des Objektives durch ein zusätzliches XY-Achsensystem auf der Probe verschoben. Mit dieser Kombination von Achsensystem und Galvoscanner können kleinere Teilstrukturen zu einer größeren Struktur zusammengefügt werden, was die Herstellung großflächiger Strukturen mit hohen Schreib-geschwindigkeiten ermöglicht.<br />Diese Arbeit befasst sich mit der Herstellung von großflächigen Membrane (5x5mm²), die für die Untersuchung von CaCo-2 Zellen verwendet werden. Die Membranporosität soll dafür bei gleichbleibender Porengröße im Bereich von 10 bis 60 % wählbar sein. Dafür wurden 3 verschiedene Membrantypen hinsichtlich ihrer Herstellungszeit und Porosität verglichen.<br />

Two-Photon polymerization (2PP) is a technique to fabricate three dimensional structures with a high spatial resolution in the range of 100nm.<br />Unfortunately the structuring speed of conventional systems for large scale structures is rather slow (10 mm/s) - resulting in a long fabrication time. By using a galvo scanner the structuring speed can be increased up to 500mm/s; but the lateral dimension of the structures is limited by the field of view of the focusing optic. Combining the galvo scanner with a XY stage extends the possible lateral size and allows the fabrication of large structures at high speed by stitching of smaller parts. This work deals with the fabrication of membranes with a large lateral size of 5x5mm². These structures are used to investigate the influence of the membrane porosity on the growth of CaCo-2 cells and therefore have to be scaled in the range from 10 to 60% - without changing the pore size. Three different types of membrane designs are tested and compared regarding structuring time and porosity.
