Die enantiospezifische enzymatische 1,2-Dihydroxylierung von Natriumbenzoat repräsentiert eine Umsetzung bei der ein sehr vielseitiges Produkt, welches auf klassisch chemischen Weg nicht zugänglich ist, für weitere synthetische Umsetzungen gewonnen werden kann.Die Möglichkeit des Up-scalings dieser enzymatischen 1,2-Dihydroxilierung wurde untersucht und nach der Optimierung des Fermentationsprozesses konnten Multigramm Quantitäten des erwarteten chiralen 1,2-dihydroxylierten Produktes für weiter synthetische Umsetzungen hergestellt werden. Die Verwendung von 1,2-dihydroxyliertem Natriunbenzoat in der organischen Synthese konnte anhand von intramolecularen Diels-Alder Reaktionen, welche zu verschiedenen hetero- und carbo-bicyclischen Verbindungen führten, gezeigt werden. Diese bicyclischen Verbindungen repräsentieren synthetische Analoga des Naturstoffes Penicillon A. Uvarirufol A, ein polyoxygenierter Naturstoff, welcher aus Uvaria Rufa isoliert wurde, wurde erstmalig versucht zu synthetisieren, ausgehend von enymatisch 1,2-dihydroxyliertem Natriunbenzoat.
The enantiospecific enzymatic 1,2-dihydroxylation of sodium benzoate represents a powerful methodology to obtain a very interesting synthon for organic synthesis due to the fact that this transformation cannot be achieved with classical oxidation reagents, in particular in asymmetric mode.The possibility of up-scaling of the enzymatic 1,2-dihydroxylation was elaborated and after optimization of the fermentation process multi-gram quantities of the expected chiral 1,2-dihydroxylated compound became available for subsequent synthetic elaboration. Application of the dihydroxylated sodium benzoate to organic synthesis is outlined towards various hetero and carbo bicyclic compounds obtained from intramolecular Diels-Alder cyclization reactions which would lead to synthetic scaffolds as structural analogs of the natural product penicillone A. Uvarirufol A, a polyoxygenated cyclohexen natural product, which was first isolated from Uvaria rufa, was attempted to be synthesized for the first time using 1,2-dihydroxilated sodium benzoate as starting material.
