Organophosphates (OP) are prevalent components in pesticides and chemical weapons, such as Malathion or Sarin gas [1]. As Acetylcholine esterase inhibitors, exposure to OPs has been proven to be fatal or to cause severe neurological disorders [1,2]. Therefore, regular environmental monitoring to detect OP concentrations is essential in agricultural applications and needed for the military sector to ensure the safety of personnel and civilian populations [2,3]. State-of-the-art analytical detection methods such as mass spectroscopy and liquid chromatography require time-consuming pre-treatments, costly reagents and are not suited for in-field usage [1,3]. Furthermore, current detection methos need to be calibrated, are specific to a single toxic compound and fail to account for the complex interactions within the human body. There is an urgent need for enhanced methods to assess environmental organophosphate neurotoxicity without endangering human or animal lives [2]. This study developed a multi-organ microphysiological system for in-field deployment capable of acute neurotoxicity evaluation. This Body-On-A-Chip model consists of five interconnected organ models and two micropumps for increases organ-to-organ interactions and media distribution. The model mimics the pathway of OPs through the skin into the body where it interacts with the liver, kidneys and the vascular system before breaching the blood brain barrier and finally arriving at the midbrain. In order to detect the damage severity of a tested compound, the system analyses the oxygen consumption in the area around the midbrain model and relates it to its viability.
