Abstract:
Einzelmolekül-Lokalisierungsmikroskopie (SMLM) ermöglicht die Betrachtung biologischer Proben mit Auflösungen unterhalb der Beugungsgrenze. Die Implementierung von SMLM bei kryogenen Temperaturen bringt zusätzliche Vorteile mit sich, so verbessert sie die Photostabilität, vermeidet chemische Fixierung und eröffnet die Möglichkeit zur korrelativen Kryo-Licht- und Elektronenmikroskopie. Gleichzeitig ist Kryo-SMLM mit neuen Herausforderungen verbunden, allen voran die anisotrope Abstrahlcharakteristik von Fluorophoren mit fester Dipolorientierung. Die sich daraus ergebenden Lokalisierungsfehler können durch Einbeziehung von Schätzungen der Dipolorientierung in das Punktspreizfunktionsmodell korrigiert werden. In dieser Arbeit wird die experimentelle Umsetzung eines in ein Kryo-SMLM-Setup integrierten Biplane-Imaging-Systems zur Bestimmung der Dipolorientierung vorgestellt. Für die Validierung des Systems wurde Polarisationsmodulation als unabhängige Referenzmethode implementiert. Es wurden Messungen an fluoreszenzmarkierten DNA-Origami-Proben durchgeführt und die bestimmten Azimutalwinkel verglichen. Während keine quantitative Übereinstimmung der beiden Methoden erzielt werden konnte, wurden experimentelle und analytische Herausforderungen identifiziert, die Ansätze für zukünftige methodische Verbesserungen liefern.

Single-molecule localization microscopy (SMLM) enables imaging of biological samples at resolutions below the diffraction limit. Limitations of room-temperature imaging have motivated the implementation of SMLM under cryogenic conditions. cryoSMLM enhances photostability, avoids chemical fixation and offers the potential for correlative imaging with cryo-electron microscopy. Nevertheless, the method introduces new complexities, most notably the need to account for the anisotropic emission characteristics of fluorophores with fixed dipole orientations, which otherwise result in significant localization bias. By incorporating estimates of the molecular dipole orientations into the point spread function model, bias-free localization can be achieved. This work presents the experimental implementation of a biplane imaging system for dipole orientation estimation, integrated into a cryo-SMLM setup. To assess the performance of the system, polarization modulation was implemented as an independent reference method. Measurements were performed on fluorescently labeled DNA origami samples, enabling a comparison of azimuthal angle estimates. While no quantitative agreement between the two methods could be achieved, experimental and analytical challenges were identified, providing insights for future methodological improvements.
