Abstract:
Phosphatidylinositolphosphate (im Folgenden als PIP bezeichnet) sind wichtige Vertreter der Klasse inositolenthaltender Naturstoffe und nehmen einen essenziellen Platz innerhalb der hormonabhängigen Signaltransduktionspfade und anderer zellulärer Regulationsmechanismen ein. Aufgrund seiner Rolle als Ausgangsstoff für die Produktion des 2nd-messenger Inositol-1,4,5-triphosphat (IP3), stellt Phosphatidylinositol-4,5-bisphosphat (PI-4,5-P oder PIP2) den wichtigsten Vertreter der obengenannten Klasse dar. IP3 ist befähigt, nach seiner Freisetzung Ca2+-Kanäle am Endoplasmatischen Retikulum (ER) zu öffnen und dadurch das Einströmen von Ca2+-Ionen in das Cytoplasma der Zelle zu bewirken. Dadurch wird das Protein Calmodulin rekrutiert, welches seinerseits an einer Vielzahl von zellulären Regulationsmechanismen beteiligt ist. 2012 wurde in der Arbeitsgruppe um Harald Sitte entdeckt, dass PI-4,5-P eine bestimmende Rolle im Oligomerisierungsverhalten des Serotonintransporters (SERT) spielt, was durch Arbeiten der Schütz-Gruppe zusätzlich zementiert wurde. Hierbei wurde mithilfe der von ihnen entwickelten fluoreszenzmikroskopischen Methode „Thinning Out Clusters While Conserving Stoichiometry Of Labelling“ (TOCCSL) aufgeklärt, dass SERT in Nähe des ER, des Ortes seiner Biosynthese, dazu neigt Oligomere, die aus einer geringen Anzahl von Untereinheiten bestehen, zu formieren. In starkem Kontrast hierzu neigt SERT an der Zellmembran, am Ort seiner biologischen Wirkung, zur Ausbildung von stabilen Oligomeren, welche aus einer Vielzahl von Untereinheiten bestehen. Um nun die Wechselwirkungen zwischen SERT und PIP mithilfe fluoreszenzmikroskopischer Methoden genauer untersuchen zu können, bedarf es eines PIP-Analogons, welches die Fähigkeit besitzt einen Fluoreszenzfarbstoff zu binden. Diese Arbeit beschäftigt sich mit der Etablierung eines Pfades innerhalb der Mihovilovic-Forschungsgruppe, der zur chemischen Synthese eines solchen Moleküls (1) führt. Hierbei wurden Methoden der Desymmetrisierungsund Schutzgruppenchemie an myo-inositol und des systematischen Aufbaus von Lipiden erforscht, wobei die Synthese der Bausteine 1 und 2 erfolgte. Die im Zuge dessen etablierten Arbeitstechniken werden in naher Zukunft zur Synthese von hochfunktionalisierten PIP-Analoga beitragen.

Phosphatidylinositolphosphates (from here on referred as PIPs) are a class of inositol-containing natural products that play crucial roles in hormone mediated signal transduction pathways and other cellular regulatory processes. Among all naturally occurring PIPs, phosphatidylinositol-4,5-bisphosphate (PI-4,5-P or PIP2) holds a special place, as it is the precursor of the 2nd messenger inositol-1,4,5-triphosphate (IP3), which plays a major role in the liberation of Ca2+ from the endoplasmatic reticulum (ER). The resulting increased concentrations of Ca2+ in the cytosol lead to the activation of calmodulin, which is involved in a magnitude of cellular regulation mechanisms. In 2012 Sitte et al. found evidence that, apart from being involved in the above mentioned processes, PIP2 regulates the oligomerization of the serotonine-transporter (SERT). This was proven by Schütz et al. using the “Thinning Out Clusters While Conserving Stoichiometry of Labeling” (TOCCSL) method, which was established in the formers lab. Through GFP-labelling it was shown that SERT at the ER, the site of its formation, tends to form oligomers composed of small numbers of monomers. In addition, rapid monomer exchange was observed, whereas SERT-oligomers at the cell membrane, their site of action, showed no observable exchange and are composed of a larger number of monomers. To further elucidate the process of SERT-oligomer assembly via fluorescence microscopy, a photo-clickable probe is needed, resembling a natural PI. In this thesis, the pathway towards the synthesis of such a probe (1) is presented. In a proof-of-concept approach, the emphasis was put on establishing the synthetic toolbox for the assembly of the structural building blocks leading to compound (1) within our lab. Therefore, methods for selective myo-inositol desymmetrization/protection and lipid-assembly were studied and culminated in the synthesis of Building Block 1 and 2. The gained insights into these types of chemistry will empower fast and concise synthesis of scaffolds resembling natural PIP to a larger extent, combined with a higher degree of functionalization, in the near future.
